ABSTRACT

Objective@#To explore effectiveness of exercise intervention and group cognitive therapy on mobile phone dependence in college students.@*Methods@#A total of 762 college students from a university in Wuhan were selected through random cluster sampling method in Oct. 2019. Among them, 126 students with higher risk for mobile phone dependence were selected as experimental group which were assigned to four groups:open exercise group(basketball,31),close exercise group(baduanjin,31), group cognitive group(30) and control group(34). The exercise group attended 60 min intervention twice a week for 10 weeks. The effectiveness was evaluated through the changes in the Mobile Phone Addiction Index (MPAI).@*Results@#The 10week exercise and group based cognitive therapy intervention can improve mobile phone dependence behavior among college students ( F group= 4.54, F time = 3.67, P <0.05). Among them, the open exercise group and the group cognitive therapy group showed slightly better effects( t= 0.06, P >0.05). In the follow up visit two months after intervention, no significant difference between the closed exercise group and the post test results( t=1.23, P >0.05). In the dimensions of out of control and withdrawal, the open exercise was more effective than the closed exercise intervention( t=2.58, 2.67, P <0.05).@*Conclusion@#Open exercise group has similar effect on improvement in mobile phone dependence with group cognitive therapy among college students, while closed exercise intervention shows better long term effect.

ABSTRACT

Objective To observe the effects of curcumin on the lung collagen area and the expression of TNF-α,IL-6 and NE in paraquat-poisoning rats at different intervals, and discuss the possible mechanism of curcumin antagonizing paraquat poisoning. Methods A total of 108 SPF Wistar rats were divided into three groups (random number):blank group (B group) for control, paraquat poisonin group (PQ group) and curcumin-treatment group (PC group). The rats of PQ group and PC group were given paraquat (50 mg/kg) by gavage, and the rats of B group were given equal volume of sterile saline solution at the same time.Thirty minutes later, the rats of PC group were given curcumin (200 mg/kg) by intraperitoneal injection, and rats of B group and PC group were given equal volume of sterile saline solution instead. At 3 d,7 d and 14 d after modeling, the distribution and pathological changes of lung tissue and collagen fiber were observed by HE and Masson staining. The concentration of tumor necrosis factor (TNF-α) and interleukin-6 (IL-6) were detected by enzyme-immunoassay.The lung neutrophil elastase (NE)expression was observed by immuno-histochemical method.Result Compared with B group,PQ group had pulmonary alveolitis in different degrees at different intervals, and the most serious pulmonary alveolitis was observed at 7 d after modeling.Diffused pulmonary fibrosis of the lung tissue and a large area of collagen fiber deposition were observed especially at 14 d after modeling,as well as the expression of NE was observed obviously, especially at 14 d after modeling. The concentration of TNF-α,IL-6 in serum were significantly increased (P < 0.05,P < 0.01).Compared with PQ group, the pulmonary alveolitis and fibrosis obviously in PC group with obvious reduction in the expression of NE and significant descrease in the concentrations of TNF-o and IL-6 (P < 0.05).Conclusion Inhabiting inflammatory factors to alleviate the seriousness of alveolar inflammation and pulmonary fibrosis might be one of the mechanism of treatment with curcumin for paraquat poisoning rats.

ABSTRACT

Objective To observe the effects of curcumin on pulmonary fibrosis and functions on paraquat (PQ)-challenged rats, and investigate the possible mechanism. Methods 108 SPF Wistar rats were divided into three groups according to random number sheet: normal saline (NS) control group, PQ model group and curcumin-treatment group. The rats in each group were subdivided into three subgroups according to different time points (3, 7, 14 days), with 12 rats in each subgroup. PQ-challenged models were reproduced by intragastrical administration of PQ solution 50 mg/kg, and those in NS control group were given the equal volume of NS. After 30 minutes, the rats in curcumin-treatment group were given 200 mg/kg of curcumin by intraperitoneal injection, and those in NS control group and PQ model group were given the equal volume of NS. At 3, 7, 14 days, the tidal volume (VT) was examined, and the blood was drawn from femoral artery for blood gas analysis. Then the rats were sacrificed and the lung tissues were harvested, the hydroxyproline (Hyp) content was measured by alkaline hydrolysis; the expression of transforming growth factor-β1 (TGF-β1) was determined by immuno-histochemistry; the distribution and the change of the pulmonary collagen fiber were observed after Masson staining. Results After exposure to PQ, the VT and arterial partial pressure of oxygen (PaO2) were decreased gradually, and the levels of Hyp and TGF-β1 were increased gradually, reaching the trough or the peak at 14 days, which were significantly lower or higher than those in NS control group [14-day VT (mL):1.52±0.33 vs. 2.81±0.47, 14-day PaO2(kPa): 5.87±0.95 vs. 14.15±1.02, 14-day Hyp (μg/mg): 3.12±0.06 vs. 1.14±0.05, 14-day TGF-β1 (integral A value): 29.72±4.27 vs. 4.15±0.52, all P < 0.01]. After intervene of curcumin, the parameters were significantly improved as compared with those of PQ model group [14-day VT (mL): 2.34±0.19 vs. 1.52±0.33, 14-day PaO2(kPa): 10.23±1.01 vs. 5.87±0.95, 14-day Hyp (μg/mg): 2.31±0.04 vs. 3.12±0.06, 14-day TGF-β1 (integral A value): 15.46±2.89 vs. 29.72±4.27, all P < 0.01]. It was shown by Masson staining that in PQ model group, with the PQ-poisoned time prolonging, diffused pulmonary fibrosis and a large number of collagen deposition were observed gradually, and the most serious collagen deposition was observed at 14 days; after intervene of curcumin, pulmonary fibrosis was alleviated significantly at different time points as compared with the PQ model group. Conclusion Curcumin can enhance the pulmonary function by reducing the deposition of collagen fiber and inhabiting pulmonary fibrosis of PQ-poisoned rats.

ABSTRACT

Objective To study the anti-tumor effect of tenacissoside H on Lewis lung cancer mice, and explore its impacts on immune function of tumor-bearing mice. Methods Mice was injected Lewis lung cancer cells subcutaneously to the right axilla. Thirty successful tumor-bearing mice were randomly divided into model group, the cyclophosphamide (CTX) group and tenacissoside H group, 10 mice in each group. Three days after inoculation, mice were intraperitoneally injected by normal saline, CTX and tenacissoside H respectively every two days, 0.2 mL in each mouse. On the 21st day, the eyeballs were extracted and blood was drawn, tumor tissue, spleen and thymus were taken and weighted to calculate tumor inhibition rate, spleen index and thymus index, and the contents of IL-2 and IL-10 were detected by ELISA. Results The tumor weights of CTX group and tenacissoside H group were lower than that of the model group with significant difference (P<0.05), and the tumor inhibition rates were 54.12%, 25.68%. The thymus index and spleen index of tenacissoside H group increased, but that of CTX group decreased significantly. Compared with the model group, the IL-2 level of tenacissoside H group was significantly increased, while the IL-10 level decreased. Conclusion Tenacissoside H can inhibit growth and metastasis of Lewis lung cancer, regulate the expression of IL-2 and IL-10, and improve the immune function of tumor-bearing mice.

ABSTRACT

<p><b>OBJECTIVE</b>To study the relationship between glutathione S-transferase M1 (GSTM1) genotypes and lipid peroxidation of asbestos workers.</p><p><b>METHODS</b>94 asbestos workers and 51 controls were selected as subjects. The general information, occupational history and individual habits were collected by questionnaires in all participants. The venous blood was sampled and the plasma was separated for the detection of malondialdehyde (MDA) level and lymphocytes for DNA isolation and GSTM1 genotyping.</p><p><b>RESULTS</b>MDA level was significantly higher in asbestos workers [(0.283 +/- 0.054) nmol/L] than that in controls [(0.163 +/- 0.053) nmol/L, P < 0.01], however, neither duration of exposure nor accumulated asbestos exposure dose was related to MDA levels; MDA levels in control workers with GSTM1 +/- genotype [(0.190 +/- 0.034) nmol/L] were significantly higher than that in control workers with GSTM1 +/+ genotype [(0.138 +/- 0.055) nmol/L, P < 0.01]. Among asbestos workers, the same trend could be found, but the differences was not significant(P > 0.05). When the workers were stratified by duration of exposure or accumulated asbestos exposure dose, MDA levels in individuals with GSTM1 -/- genotype were also higher than those with GSTM1 +/+ genotype, but the differences were also not significant(P > 0.05).</p><p><b>CONCLUSION</b>Both exposure to asbestos and deficiency of GSTM1 genotype were related to lipid peroxidation in workers, but the role of the former may be more important than that of the latter.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To understand the effects of exposure to asbestos and GSTM1 genotypes on plasma activity of glutathione S-transferases (GSTs).</p><p><b>METHODS</b>Ninety-four workers exposed to asbestos and 51 controls were selected, and their general information, occupational history and personal behavior were collected by questionnaire. Venous blood specimen was collected from each of them and plasma was separated for detection of GSTs activity and lymphocytes for DNA extraction and GSTM1 genotyping.</p><p><b>RESULTS</b>Plasma activity of GSTs in the asbestos-exposed workers (23.0 +/- 6.9) U/L was significantly lower than that in the controls (32.6 +/- 11.8) U/L, which declined with the length of employment in asbestos industry and the increase of cumulated dose of asbestos. Stratification of workers by GSTM1 genotypes showed that plasma activity of GSTs in asbestos-exposed workers with GSTM1+/+ or GSTM1-/- were (24.0 +/- 6.1) and (22.5 +/- 7.3) U/L, respectively, lower than those in the controls with the same genotypes (38.1 +/- 13.2) and (26.8 +/- 6.6) U/L. Plasma activity of GSTs in the control workers with GSTM-/- was significantly lower than in those with GSTM+/+, and, so did in asbestos-exposed workers, but without statistically significant difference.</p><p><b>CONCLUSION</b>Exposure to asbestos could significantly decrease plasma activity of GSTs, and GSTM1 genotypes could affect on the activity of GSTs in the control workers, which was not so obvious in asbestos-exposed workers.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effects of chrysotile on the activities of some enzymes for xenobiotics metabolism.</p><p><b>METHODS</b>UICC chrysotile (UC) and China Mangya chrysotile (MC) asbestos fibers were used at different doses (0, 25, 50, 100, 200 mg/L) to study its effects on the activities of cytochrome P4501A1 and glutathione S-transferase (GST) in A549 cell line. Also, the effects of chrysotile on the activities of CYP1A1 and GST induced by benzo(a)pyrene were studied.</p><p><b>RESULTS</b>The activity of EROD increased slowly with the increasing dose of UC, as A549 cells were incubated with UC for 24 h, and EROD activity increased by 40% at a dose of 200 mg/L UC. However, activity of EROD decreased by 32% with 48 h incubation at the same dose, indicating that lower dose of UC and short time could induce the activity of EROD in A549 cells, whereas higher doses and long time could inhibit its activity. MC exhibited a multiphasic effects on the activity of EROD, whether at a dose of 25 mg/L for 24 h or 48 h all gave the strongest induction, 1.86 times or 1.28 times as controls, respectively. However, EROD activity decreased with the increases in MC doses and incubation time, with the lowest as 35% of the controls. The effect of UC on GST activity was not so obvious, with the highest as the increase in its activity by 20%. The highest induction of MC to GST activity was at a dose of 25 mg/L, 2, 5 times as that in controls. With the increases in its doses, effects of MC on GTS activity became inhibition from induction, like that on EROD activity. MC at a dose of 200 mg/L could lower the activity of GST by 18.7%. A549 cells were incubated with chrysotile fiber for 24 h firstly, and then incubated with benzo(a)pyrene to induce the activities of EROD and GST. The results showed that neither UC nor MC could affect the activity of EROD induced by benzo(a)pyrene. However, UC at a dose of 200 mg/L and MC at 100 mg/L could increase the activity of GST induced by benzo(a)pyrene.</p><p><b>CONCLUSION</b>Chrysotile at different doses or its different types showed varied effects on the activities of EROD and GST in A549 cell lines, probably because of different physicochemical characteristics and surface activity of two kinds of asbestos.</p>
